SBF - SEM

Serial block face scanning electron microscopy (SBF-SEM) enables high resolution 3D imaging of biological samples that have been fixed, stained and embedded in resin. SBF-SEM provides 5-10 nanometer resolution of the ultrastructure over relatively large fields of view (typically 25 x 25um) in the x,y of the section plane – and > 25nm resolution over hundreds or thousands  of microns depth (z or sectioning direction).

An SBF-SEM consists of an ultramicrotome installed inside an SEM. The SEM collects sequential images of the block face while the ultramicrotome incrementally removes surface sections. The sequential  images can be segmented and reconstructed to provide 3D data.

A BBSRC ALERT17 grant funded SBF-SEM (Zeiss Gemini SEM 450/Gatan 3View) was installed in the Wolfson Bioimaging Facility in February 2019.
